Identification of Sugarcane S. spontaneum (Poaceae) Germplasm: Evidence from rDNA-ITS and rDNA Locus Analyses

Sugarcane is a major crop for sugar production around the world. The complexity of sugarcane genome creates challenges use both conventional and molecular breeding methods to improve at genetic level. DNA sequencing an important tool assess how genus Saccharum genera complex are interrelated. Here, we identify kinship Nepal2013-6 (Saccharum spontaneum, x = 10) using tetra-primer amplification refractory mutation system (ARMS) PCR. Based on rDNA-ITS sequence analysis, accession falls within single cluster with S. spontaneum (Yunnan82-114 SES208), which consistent previous results. Moreover, fluorescence in situ hybridization (FISH) results indicate that 5S rDNA spots chromosomal ploidy analytical materials, whereas 35S has similar or fewer sites than ploidy. Therefore, FISH patterns would be more suitable analysis varied basic chromosome number 8, 9, 10. Altogether, these sequences will useful marker further rapidly identifying relationship breeding.
